The eosin test marks dead spermatozoa, with damaged membrane, violet/purple. On the other hand, live spermatozoa remain white, since their membrane is intact and cannot penetrate the dye.
The man should obtain the semen sample by masturbation, maintaining a period of abstinence of approximately 3-5 days for the results to be reliable.
