One test used to evaluate the level of sperm DNA fragmentation is the Sperm Chromatin Dispersion (SCD) test, which involves the decondensation of the sperm chromosomes in order to prevent DNA loops from forming.
As a consequence, sperms with intact DNA will have large halos of dispersed DNA, while sperms with fragmented DNA won't expand equally and will form smaller halos or even no halos at all.
